Interferon-γ Regulates Cellular Metabolism and mRNA Translation to Potentiate Macrophage Activation
Scientists found that interferon-γ regulated the metabolism and mRNA translation of human macrophages by targeting the kinases mTORC1 and MNK, both of which converge on the selective regulator of translation initiation eIF4E. [Nat Immunol] Abstract

Epigenetic Therapy Overcomes Treatment Resistance in T Cell Prolymphocytic Leukemia
Investigators report seven complete and one partial remission in eight patients receiving alemtuzumab and cladribine with or without a histone deacetylase inhibitor. Monozygotic Twins Discordant for Common Variable Immunodeficiency Reveal Impaired DNA Demethylation during Naïve-to-Memory B-Cell Transition
The authors performed a high-throughput DNA methylation analysis of common variable immunodeficiency (CVID) using a pair of CVID-discordant MZ twins and showed predominant gain of DNA methylation in CVID B cells with respect to those from the healthy sibling in critical B lymphocyte genes, such as PIK3CD, BCL2L1, RPS6KB2, TCF3 and KCNN4. Metabolic Plasticity of Human T Cells: Preserved Cytokine Production under Glucose Deprivation or Mitochondrial Restriction, but 2-Deoxy-Glucose Affects Effector Functions
Investigators analyzed glycolytic and mitochondrial activity in correlation to function in activated human CD4 and CD8 T cells. Glycolysis was barely detectable upon stimulation but accelerated beyond 24 hours, whereas mitochondrial activity was elevated immediately in both T-cell populations. [Eur J Immunol] Abstract

The BMP Pathway Participates in Human Naive CD4+ T Cell Activation and Homeostasis
Scientists demonstrated that canonical bone morphogenetic protein (BMP) signaling is necessary during two critical events that regulate the size and function of human naive CD4+ T cell population: activation and homeostasis. [PLoS One] Full Article

Development of Depressive Symptoms Post Hip Fracture Is Associated with Altered Immunosuppressive Phenotype in Regulatory T and B Lymphocytes
Researchers assessed the frequency of regulatory T cells and IL10 production by CD4 T cells, and the frequency and IL10 production by regulatory B cells in 101 hip fracture patients six weeks after injury and 43 healthy age-matched controls. [Biogerontology] Full Article

B Cell-Derived Circulating Granzyme B Is a Feature of Acute Infectious Mononucleosis
Researchers showed that patients with recently diagnosed infectious mononucleosis caused by Epstein-Barr virus (EBV) have high circulating levels of granzyme B (GzmB) that may be derived from infected B cells early in course of disease. They found that infecting B cells with EBV greatly amplified GzmB secretion in response to the same stimuli, but the expression was terminated once the infection had become latent.
